Abstract

The low production of orchids in Indonesia is mainly due to the limitations of conventional propagation methods, which are time-consuming and result in low quantities of plantlets. This research aimed to determine the interaction between Benzyl Amino Purine (BAP) and Naphthalene Acetic Acid (NAA) and to obtain the best combination for the growth of Dendrobium Gradita 31 seedlings. The experiment was conducted from May to June 2025 at the Plant Tissue Culture Laboratory, Politeknik Negeri Lampung, using a factorial randomized block design (RBD) with twelve combinations of BAP (0, 1, and 2 mg/l) and NAA (0, 0.5, 1, and 1.5 mg/l). The observed parameters included seedling height, number of leaves, root length, number of roots, and seedling weight. The results showed a significant interaction between BAP and NAA on all observed parameters. The best combination for seedling height was obtained at 1 mg/l BAP + 1.5 mg/l NAA, while the highest number of leaves was achieved with 1 mg/l BAP + 0 mg/l NAA. The best root length was found at 0 mg/l BAP + 0.5 mg/l NAA, and the highest number of roots occurred at 0 mg/l BAP + 1.5 mg/l NAA. The combination of 0 mg/l BAP + 0 mg/l NAA produced the heaviest seedling weight. These results indicate that appropriate concentrations and interactions of BAP and NAA can significantly enhance the growth performance of Dendrobium Gradita 31 seedlings under in vitro conditions.
